Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
GAD65 (glutamic acid decarboxylase-65)는 자가면역질환(제1형 당뇨병, stiff man syndrome, 다내분비 자가면역질환 등)에서 발견됩니다.
GAD65에 대한 자가항체는 새로 진단된 제1형 당뇨병 환자의 약 60–70%에서 발견되며, 질병 활성의 중요한 지표로 사용됩니다.
이 항체들은 주로 GAD65의 구조 의존적 영역을 인식하며, 다른 동형체인 GAD67에는 거의 결합하지 않습니다. GAD67에 대한 자가항체는 약 15%의 초기 제1형 당뇨병 환자에서만 발견되며, 대부분의 결합은 GAD65로 차단될 수 있습니다. 이는 두 동형체 간의 공유된 에피토프를 시사합니다.
