Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at room temperature for 20 min.
Target Information
GAD65 (glutamic acid decarboxylase-65)는 자가면역 질환, 특히 인슐린 의존성 제1형 당뇨병(IDDM), stiff man syndrome, polyendocrine autoimmune disease 등에서 발견됩니다.
GAD65에 대한 자가항체는 새로 진단된 IDDM 환자의 60–70%에서 발견되며 질병 활성도의 중요한 지표로 사용됩니다.
이 항체들은 주로 GAD65의 입체 구조 의존적 영역을 인식하며, GAD67에는 거의 결합하지 않습니다.
GAD67에 대한 자가항체는 최근 발병한 IDDM 환자의 약 15%에서만 발견되며, 대부분 GAD65에 의해 결합이 차단되어 두 isoform 간의 공유 epitope 존재를 시사합니다.
