• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature (RT).
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at RT for 20 minutes.

Target Information

GAD65 (glutamic acid decarboxylase-65)는 인슐린 의존성 당뇨병(IDDM), 강직인 증후군(stiff man syndrome), 다선성 자가면역 질환(polyendocrine autoimmune disease) 등 자가면역 질환에서 발견됩니다.
GAD65에 대한 자가항체는 신규 IDDM 환자의 60–70%에서 검출되며, 질병 활성의 중요한 지표로 사용됩니다.
이 항체들은 주로 GAD65의 입체 구조 의존적 부위를 인식하며, GAD67에는 거의 결합하지 않습니다.
GAD67에 대한 자가항체는 최근 발병한 IDDM 환자의 약 15%에서 발견되며, 대부분 GAD65에 의해 결합이 차단되어 두 아이소폼 간의 공통 에피토프 존재를 시사합니다.
