Target Information
HIF1AN (Hypoxia Inducible Factor 1 Subunit Alpha Inhibitor) 단백질은 HIF-1α의 C-terminal transactivation domain (CAD) 내 Asn-803 잔기를 수산화화하여 산소 감지 기능을 수행합니다. 정상 산소 조건에서는 이 수산화 반응이 HIF-1과 전사 보조인자(Cbp/p300-interacting transactivator)의 결합을 억제합니다. 또한 HIF1A, VHL, 히스톤 탈아세틸화효소(HDACs)와 상호작용하여 전사 억제에 관여합니다.
HIF1AN은 NFKB1, NFKBIA, NOTCH1, ASB4, PPP1R12A 등 여러 ankyrin repeat domain(ARD) 단백질의 특정 Asn 잔기를 수산화화하며, ANK1과 TNKS2의 Asp 및 His 잔기도 수산화합니다.
이 단백질은 NOTCH1 활성을 음성 조절하여 근육 분화를 촉진하고, ASB4 활성을 양성 조절하여 혈관 분화를 촉진합니다.
