The monoclonal antibody IM348 recognizes mouse CD6, a transmembrane glycoprotein of the scavenger receptor family.
CD6 contains three SRCR domains, a transmembrane region, and a variable cytoplasmic tail depending on isoform. It is expressed on most T lymphocytes. In mice, B cells in the spleen lack CD6, unlike humans where a subset of B lymphocytes express it.

The CD6 ligand is CD166 (ALCAM), involved in adhesion and migration during organogenesis. The intracellular domain of CD6 is constitutively phosphorylated, suggesting a role in T cell activation and signaling. In humans, depletion of CD6-expressing cells increases graft-versus-host tolerance in transplants.

Applications Tested: Flow cytometric analysis of mouse splenocytes. Recommended ≤0.25 µg/test (100 µL final volume).

Target Information

CD6 (T12) is a member of the scavenger receptor superfamily expressed on T and B cell subsets, thymocytes, and acute lymphocytic leukemia (ALL) cells.
It interacts with CD166/ALCAM, acting as a coreceptor for T cell activation and stabilizing the immunological synapse. CD6–ALCAM mediated adhesion supports T cell proliferation.
CD6 may associate with CD5 to fine-tune its signaling. Ligation of CD6 has an anti-apoptotic role in chronic lymphocytic leukemia B cells.
