Product Specific Information

The D7 monoclonal antibody reacts with mouse Sca-1, an 18 kDa member of the Ly-6 family of GPI-linked surface proteins.
D7 binds both Ly-6E.1 and Ly-6A.2 molecules expressed by mouse hematopoietic stem cells, myeloid populations, and peripheral T and B cells.
Non-activated splenocytes from Ly-6.2 strains (C57BL, SJL, 129, AKR, etc.) show higher frequency of D7-positive cells compared to Ly-6.1 strains (BALB/c, C3H, NZB, etc.).
Sca-1 is expressed at high levels upon activation regardless of Ly-6 haplotype and is a major phenotypic marker for mouse hematopoietic progenitor/stem cell subsets.

Applications Tested: Flow cytometric analysis of mouse bone marrow cells (≤0.5 µg/test).
A test is defined as the amount of antibody that stains a cell sample in 100 µL volume.
Cell number may vary (10⁵–10⁸ cells/test); antibody should be titrated for optimal performance.

Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation; protect vial and samples from light.

Fixation: Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)
  Minimal impact on brightness and FRET efficiency/compensation. Clone-specific performance should be determined empirically.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Stem Cell Antigen-1 (Sca-1) is a member of the Ly-6 family, expressed on multipotent hematopoietic stem cells (HSC) and used as a marker of HSC in mice.
Sca-1 is used with lineage depletion antibodies to identify murine HSC and is found in fetal liver, adult bone marrow, peripheral blood, and spleen.
It may be involved in the regulation of B and T cell activation.
