Target Information

The protein tyrosine phosphatase PTPN22 (also known as PTP22, LYP, PEP, formerly PTPN8) is a genetic variant associated with increased risk of autoimmune diseases such as type 1 diabetes and rheumatoid arthritis.
A missense SNP in PTPN22 encodes a hematopoietic-specific phosphatase known as Lyp. The risk allele appears in approximately 17% of the general Caucasian population and about 28% of individuals with rheumatoid arthritis.
This variant is thought to disrupt the P1 proline-rich motif essential for interaction with the SH3 domain of CSK (cytoplasmic tyrosine kinase), potentially altering T cell activation regulation.
PTPN22 may act as an effector or regulator of CSK in T cells and other hematopoietic cells.
