Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding mature erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase enzymatic activity and plays an important role in lymphocyte activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse bone marrow cells (≤ 0.5 µg/test).
A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

Super Bright 436 is excited with the violet laser line (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells (~10% of surface proteins in lymphocytes). It is absent on non-hematopoietic cells and tissues. CD45 glycoprotein regulates lymphocyte development and antigen signaling via Src-family kinases.

CD45 exists as multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. CD45RA is one isoform with restricted expression among lymphoid subtypes. Isoforms vary in membrane localization and expression depending on cell type and physiological state.

CD45 regulates T- and B-cell receptor signaling, suppresses JAK kinases, and modulates integrin-mediated survival and apoptosis pathways. It plays a key role in immune function regulation.
