Target Information
Presenilin 1 was initially identified as a marker of susceptibility to early-onset Alzheimer’s disease.
Along with PEN2, nicastrin, and APH-1, Presenilin 1 forms the γ-secretase protein complex, a membrane-bound aspartyl protease that cleaves certain proteins within the hydrophobic environment of the lipid bilayer.
This cleavage is crucial for signaling from several cell-surface receptors and is required for generating neurotoxic amyloid peptides central to Alzheimer’s disease pathogenesis.
Similar to TACE and BACE protease families, γ-secretase cleaves the amyloid precursor protein (APP) within its intramembrane region, producing either the non-toxic p3 peptide or the toxic Aβ amyloid peptide.
Accumulation of Aβ peptide is thought to precede Alzheimer’s disease. Multiple isoforms of presenilin 1 are known to exist.
