Product Specific Information
The OKT8 monoclonal antibody reacts with the human CD8a molecule (~32–34 kDa), expressed as CD8αβ heterodimers or CD8αα homodimers on thymocytes, mature T cells, and NK cells. CD8a binds to MHC class I and associates with p56lck, playing a critical role in T-cell development and activation.
Preliminary testing shows OKT8 does not compete with clones RPA-T8 and HIT8a for binding, indicating distinct epitopes.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells (5 µL/test).
A test corresponds to staining in 100 µL volume with 10⁵–10⁸ cells.
Note: NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
CellBlox Plus Blocking Buffer (Product # C001T06F01) must be used for optimal staining.
Add 5 µL per sample before mixing with cells.
NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures, engineered for optimized fluorescence with minimal spillover.

Target Information
CD8 is a type I transmembrane glycoprotein functioning as a co-receptor for MHC class I molecules. It is expressed on T cells, NK cells, monocytes, and dendritic cells, forming CD8αβ heterodimers or CD8αα homodimers.
CD8 engagement with MHC-I/peptide complexes recruits Lck kinase, triggering cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
CTL differentiation is enhanced by IL-2, IL-12, and TGF-β1.
