Product Specific Information
The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimer or CD8αα homodimer. CD8a is found on thymocytes, mature T cells, and NK cells. It binds MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.
OKT8 does not compete with clones RPA-T8 or HIT8a for binding to human peripheral blood leukocytes, indicating distinct epitopes.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for use at 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
NovaFluor Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
Blocking Buffer Recommendation:
Use CellBlox Plus Blocking Buffer (Product # C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family, essential for T-cell signal transduction, differentiation, and activation. CD8 acts as a co-receptor for MHC class I molecules and is expressed on T cells, NK cells, monocytes, and dendritic cells. Upon MHC-I/peptide binding, CD8 recruits Lck kinase, leading to cytokine production and cytotoxic T lymphocyte (CTL) activation, crucial for pathogen and tumor clearance.
