Target Information

T-cell protein tyrosine phosphatase (TC-PTP), also known as PTPT and PTPN2, is an enzyme that removes phosphate groups covalently attached to tyrosine residues in proteins. This enzyme has two C-terminal splice variants with different subcellular localizations.

• The 45 kDa isoform is nuclear in resting cells but redistributes to the cytosol upon stimulation with growth factors and stress.
• The 48 kDa isoform is found in the endoplasmic reticulum and has distinct physiological substrates.

TC-PTP is expressed in many tissues, particularly in hematopoietic cells. Knockout mice lacking TC-PTP die 3–5 weeks after birth due to erythropoietic and lymphopoietic deficits, indicating its essential role in bone marrow maturation. TC-PTP dephosphorylates multiple phosphoproteins, including p52 Shc and receptors for EGF, insulin, and growth hormone.
