Target Information
iNOS (Inducible Nitric Oxide Synthase, NOS)는 세포 간 다양한 신호를 전달하는 무기성 기체 자유 라디칼입니다. NO 생성은 nitric oxide synthase (NOS) 계열 효소에 의해 매개되며, iNOS는 간에서 발현되고 lipopolysaccharide와 특정 cytokine에 의해 유도됩니다.
NOS는 L-arginine을 산화시켜 L-citrulline과 NO를 생성하며, 세 가지 주요 isoform(b/nNOS, eNOS, iNOS)이 존재합니다. 모든 NOS isoform은 calmodulin, NADPH, FAD, FMN 결합 도메인을 포함합니다.
iNOS는 macrophage, hepatocyte, synoviocyte, smooth muscle cell 등 다양한 세포에서 발견되며, IFN-γ, TNF, IL-1, IL-2, LPS 등에 의해 발현이 증가합니다. Cytokine 유도 후 iNOS는 지연된 활성 반응을 보이며 장기간 NO 생산이 증가합니다.
iNOS 관련 pseudogene은 염색체 17의 Smith-Magenis 증후군 영역 내에 존재하며, iNOS 기능 이상은 achalasia 및 impotence와 관련될 수 있습니다.
