Description:
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to create the IL-7 receptor (IL-7R). It is a type I glycoprotein (~75–80 kDa) expressed by immature B cells, thymocytes, and most mature T cells. IL-7 binding activates Jak/STAT pathways essential for lymphocyte development and T-cell homeostasis.

CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). Co-expression of CD4 and CD25 alone may include non-suppressive cells, but CD127 low expression enriches for Foxp3⁺ Tregs with suppressive activity.

Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product # 14-1079-80).

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.125 µg)/test.
A test is defined as the amount of antibody staining a 100 µL sample (10⁵–10⁸ cells/test).

Light Sensitivity: Protect from light (photo-oxidation sensitive).
Fixation: Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)

Excitation: 488–567 nm

Target Information

CD127 (Interleukin-7 Receptor α, IL7Rα):
A glycoprotein regulating lymphopoiesis. The IL7R complex (α/γ heterodimer) mediates IL-7 signaling through the γ(c) chain shared with IL-2R. CD127 promotes proliferation of precursor B cells, thymocytes, and T cells. Dysfunction is linked to severe combined immunodeficiency (SCID).
