Target Information

Cortactin (Cttn) is a ubiquitous actin-binding protein originally identified as a substrate for Src. It contributes to actin cytoskeleton organization and cell shape, playing a role in lamellipodia formation and cell migration. Cortactin localizes in actin-enriched peripheral structures and facilitates actin network formation.

Cortactin has four major domains:

• N-terminal acidic (NTA) domain
• Tandem repeats domain
• C-terminal proline-rich domain
• SH3 domain

The NTA domain associates with the Arp2/3 and WASP complexes at F-actin branches. Cortactin is involved in promoting cell motility and invasion, particularly in invadopodia associated with ECM degradation by cancer cells. It is phosphorylated by Src family kinases at Y421, Y466, Y482, and by Erk family kinases at S405 and S418.
