Product Specific Information

The 236A/E7 antibody reacts with human FOXP3 protein (also known as FORKHEAD BOX P3, SCURFIN, JM2). Cross-reactivity with other proteins has not been determined. FOXP3 is a 49–55 kDa transcriptional regulator, defective in scurfy mice. It is highly expressed in CD4⁺CD25⁺ regulatory T cells (Treg) and induces Treg phenotype when ectopically expressed in CD4⁺CD25⁻ cells.

Intracellular staining and flow cytometric analysis of freshly isolated human PBMCs using this antibody and the Foxp3 Staining Buffer Set (Product # 00-5523) show staining of the CD4⁺CD25^bright population.

The epitope recognized by 236A/E7 differs from that of PCH101 (Product # 72-5776-40).

Applications Tested: Pre-titrated and tested using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).

Use 5 µL (0.25 µg) per test. A test is defined as antibody amount for a 100 µL sample volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, excited by the 405 nm Violet laser and emitting at 445 nm. Ensure instrument compatibility.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is 381 amino acids long and expressed in CD25⁺CD4⁺ regulatory T cells, low in CD4⁺CD25⁻ cells, and absent in CD4⁻CD8⁺ T cells. FOXP3 acts as a master regulator and specific marker of regulatory T cells.
Mutations in FOXP3 cause the scurfy phenotype in mice and IPEX syndrome (immune dysfunction, polyendocrinopathy, enteropathy, X-linked) in humans. FOXP3 expression in cancer cells is regulated by p53 in response to DNA damage.
