Target Information

14-3-3 sigma (stratifin, SFN, epithelial cell marker protein 1) is a member of a highly conserved family of 14-3-3 proteins present in all eukaryotic organisms. There are seven known mammalian isoforms (epsilon, beta, zeta, gamma, theta, tau, sigma) that play crucial roles in cellular signaling by binding to and altering localization or stability of key molecules.
14-3-3 sigma was initially identified as a mammary epithelium marker and later recognized as a regulator of the cell cycle checkpoint. It has been reported as a prognostic marker in pancreatic cancer and is the only isoform induced by the tumor suppressor protein p53 in response to DNA damage.
14-3-3 sigma acts as a p53-regulated inhibitor of G2/M progression and functions as a tumor suppressor gene, which can be inactivated by methylation of CpG islands in epithelial tumor cells. Two isoforms of human 14-3-3 sigma are produced by alternative splicing.
