Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in the retinal pigment epithelium (RPE) phagocytosis pathway, which is implicated in human retinal disease. It regulates physiological processes such as cell survival, migration, differentiation, and efferocytosis. Ligand binding induces autophosphorylation on its intracellular domain, enabling downstream signaling through GRB2 or PLCG2 and phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1.
MERTK signaling contributes to macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and engulfment. In RPE, it regulates rod outer segment fragment phagocytosis and inhibits Toll-like receptor (TLR)-mediated innate immune response by activating STAT1, which induces SOCS1 and SOCS3.
