This antibody reacts with an extracellular class II epitope on CD34, a 110–115 kDa monomeric transmembrane phosphoglycoprotein expressed on hematopoietic progenitor cells and pluripotential stem cells. It is gradually lost on progenitor cells and can also be used as an endothelial marker.
Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It functions as a stem cell marker and may act as a receptor involved in adhesion, differentiation, and proliferation of hematopoietic stem cells. CD34 expression represents a specific developmental state of hematopoietic cells with distinct adhesion properties. It may mediate attachment of stem cells to bone marrow extracellular matrix or stromal cells and serve as a scaffold for lineage-specific glycans. The intracellular domain is phosphorylated by protein kinase C, suggesting a role in signal transduction. CD34 dysfunction is associated with diseases such as dermatofibrosarcoma and neurofibroma.
