Product Specific Information

The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. It is mainly expressed on activated T and B lymphocytes.

Two ligands, PD-L1 (B7-H1) and PD-L2 (B7-DC), interact with PD-1. J43 has been reported to inhibit the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo administration of J43 enhances contact hypersensitivity and exacerbates acute GVHD, EAE, and NOD diabetes, similar to PD-1 deficiency.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on stimulated mouse splenocytes at ≤0.5 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm and is suitable for cytometers with a 355 nm UV laser. Protect from light to prevent photo-induced oxidation.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to minimize non-specific interactions.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333). Compatible with short-term methanol-based fixation (<1 hour).

Target Information

PD-1 (programmed death-1, PDCD-1) is an inhibitory receptor expressed on activated T cells. It binds to ligands PD-L1 and PD-L2 on antigen-presenting cells (APCs), delivering inhibitory signals that regulate immune activation. PD-1-deficient mice exhibit hyperactivated immune responses and autoimmune diseases. Although PD-1 has a predicted molecular weight of 55 kDa, it often migrates at a higher molecular weight in SDS-PAGE.
