Description:
This 4B4 (4B4-1) monoclonal antibody reacts with human CD137 (also known as 4-1BB or TNFRSF9), an inducible member of the TNFR family of costimulatory molecules expressed on T cells, NK cells, dendritic cells, granulocytes, and mast cells. CD137 signaling recruits TRAF1 and TRAF2, contributing to T cell activation, survival of activated and CD8 memory T cells, and suppression of myelopoiesis and dendritic cell development. It can also promote expansion of CD4⁺CD25⁺ regulatory T cells ex vivo. The ligand 4-1BBL is found on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry on Con A-stimulated human peripheral blood cells. Recommended use: 5 µL (0.06 µg) per test.
A test is defined as the amount of antibody that stains a sample in 100 µL final volume. Cell number may vary from 10⁵ to 10⁸ cells/test.

Target Information

CD137 (TNFRSF9, 4-1BB) is a costimulatory receptor expressed mainly on activated T cells. Its ligand, 4-1BBL, is expressed on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells. CD137 signaling supports survival and expansion of activated and CD8⁺ memory T cells, while suppressing myelopoiesis and dendritic cell development. Triggering CD137 induces cytokine release that regulates peripheral monocyte survival. Soluble forms of CD137 may act as negative regulators of immune responses.
