Product Specific Information

The TL2.1 monoclonal antibody reacts with human Toll-like receptor 2 (TLR2). TLR2 is a type I transmembrane protein involved in innate immunity and inflammation, recognizing bacterial lipoproteins. It is expressed by peripheral blood monocytes and distinguishes different pathogens.

TL2.1 is a blocking antibody used to study TLR2’s role in microbial lipoprotein-induced cytokine production in human PBMCs. It can immunoprecipitate human TLR2 (~90 kDa) from PBMC and HMEC.

• Applications Tested: Pre-titrated and tested by flow cytometry on normal human peripheral blood cells
• Recommended Use: 5 µL (1 µg)/test in 100 µL final volume

Target Information

TLR2 is part of the Toll-like receptor family essential for pathogen recognition and activation of innate immunity. It acts through adaptor molecules like MyD88 and TIRAP to activate kinases and transcription factors such as PKC and NF-κB.

TLR2 forms heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins. It is highly expressed in peripheral blood leukocytes and mediates responses to Gram-positive bacteria and yeast.

Expression levels vary across cell types:

• Highest in CD14+ monocytes
• Followed by CD15+ granulocytes, CD19+ B-cells, and CD3+ T-cells

Regulation examples:

• Upregulated by: LPS, GM-CSF, IL-1, IL-10
• Downregulated by: IL-4, IFN-γ, TNF

Ten human homologs of TLRs (TLR1–10) have been identified. TLR2 is expressed in tonsils, microglia, lymph nodes, and activated B-cells in germinal centers.
