Product Specific Information

The MIH4 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the immunoglobulin superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. PD-1 is expressed predominantly on activated T and B lymphocytes.
Two members of the B7 family, PD-L1 (B7-H1) and PD-L2 (B7-DC), act as PD-1 ligands and exhibit overlapping functions. These ligands are constitutively expressed on some normal tissues and upregulated on activated antigen-presenting cells.
The MIH4 antibody recognizes a different epitope than clone J105.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry using stimulated normal human peripheral blood cells. Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume).
Excitation: 488 nm; Emission: 519 nm; Laser: Blue Laser.


Target Information

Cell-mediated immune responses are initiated by T lymphocytes stimulated by peptides bound to MHC molecules on antigen-presenting cells (APC). Activated T cells express PD-1 (PDCD-1), which transmits inhibitory signals upon binding to PD-L1 or PD-L2 on APCs.
PD-1 signaling downregulates immune activation in the absence of danger signals such as LPS. PD-1 knockout mice exhibit hyperactivated immune systems and autoimmune phenotypes.
Despite its predicted molecular weight, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
