Target Information
TRAIL (TNFSF10) is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells, while sparing normal cells despite its expression in most tissues. TRAIL interacts with several TNF receptor superfamily members including TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and possibly TNFRSF11B/OPG.
Binding to decoy receptors such as TRAILR3, TRAILR4, and OPG modulates its apoptotic activity. TRAIL receptor engagement activates MAPK8/JNK, caspase 8, and caspase 3 pathways. TRAIL is a type II membrane protein expressed broadly in human tissues and is known to induce apoptosis and NF-κB activation similar to TNF and Fas ligand.
