Target Information

TXK, along with close relatives BMS, ITK, and BTK, signals downstream of antigen receptors and other receptor types.
All members of the family contain SH3 and SH2 domains and, except for Txk and Dsrc28C, also contain pleckstrin homology (PH) and Tec homology (TH) domains in their amino termini.
Four alternatively spliced forms of Tec are expressed broadly in hematopoietic cells and hepatocytes.
The Emt gene product associates with CD28 and becomes activated after CD28 ligation.
Btk is essential for proper B cell development, and mutations in the Btk gene are linked to X-linked agammaglobulinemia (Bruton’s disease).
Bmx protein shares high homology with Btk and shows highest expression in the heart.
Txk expression is T cell-specific, while Drosophila Tec homolog Dsrc28C expression is developmentally regulated.
