Target Information
AHR (Ah Receptor)는 ARNT (HIF-1 Beta) 및 Drosophila 단백질 PER, SIM과 함께 단백질 패밀리에 속합니다. N-말단 약 200개의 아미노산으로 구성된 PAS 도메인을 포함하며, 다양한 조직에서 발견됩니다. AHR은 특정 DNA enhancer 서열에 결합하여 cytochrome P-450 (CYPIA1) 유전자의 mRNA 전사를 유도합니다.
AHR 유전자는 평면 방향족 탄화수소에 대한 생물학적 반응 조절에 관여하는 리간드 활성화 전사 인자를 암호화합니다. 다양한 방향족 탄화수소가 AHR의 리간드로 알려져 있으며, cytochrome P450과 같은 xenobiotic 대사 효소를 조절합니다. AHR은 다양한 척추동물 종에서 발견되는 helix/loop/helix 구조의 전사 인자입니다.
AHR의 알려진 리간드는 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)과 같은 방향족 화합물이며, 스테로이드/갑상선 호르몬 수용체와 달리 생리적 리간드는 알려져 있지 않습니다. 비활성 상태의 세포에서는 AHR이 주로 HSP90과 복합체를 이루어 세포질에 존재하며, 리간드 결합 후 핵으로 이동해 DNA 결합 형태로 변환됩니다. 이 과정은 HSP90 해리 및 Arnt 단백질과의 헤테로다이머 형성을 포함합니다.
AHR 관련 질환으로는 eosinophilic fasciitis 및 seborrheic dermatitis가 보고되어 있습니다.
