세포 내 위치

Cytoplasm and nucleus. Cytoplasmic and nuclear in the absence of TGF-beta. On TGF-beta stimulation, migrates to the nucleus when complexed with SMAD4. On dephosphorylation by phosphatase PPM1A, released from the SMAD2/SMAD4 complex and exported out of the nucleus by interaction with RANBP1. Localized mainly to the nucleus in early embryo development, later evident in cytoplasm at blastocyst and epiblast stages.

배경 정보

The protein encoded by this gene belongs to the SMAD family, which are signal transducers and transcriptional modulators mediating multiple signaling pathways. Smad2 mediates the signal of transforming growth factor (TGF)-β, regulating cellular processes such as proliferation, apoptosis, and differentiation. It is recruited to TGF-β receptors via interaction with SARA protein. Upon TGF-β stimulation, Smad2 is phosphorylated, dissociates from SARA, and associates with SMAD4—an essential step for nuclear translocation and transcriptional regulation.
