Product Specific Information
The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. It is expressed predominantly on activated T and B lymphocytes. PD-L1 (B7-H1) and PD-L2 (B7-DC) are its ligands, involved in inhibitory immune signaling.
Costaining experiments suggest that eBioJ105 recognizes a different epitope than MIH4 (Product #11-9969).
- Applications Reported: Flow cytometric analysis
- Applications Tested: Pre-diluted and tested by flow cytometry of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test.
- Laser Compatibility: Ultraviolet (355 nm) laser required for Brilliant Ultra Violet™ 615 detection.
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Target Information
T lymphocytes initiate cell-mediated immune responses when stimulated by peptides bound to MHC molecules on antigen-presenting cells (APC). PD-1 (PDCD-1) mediates inhibitory signals by binding PD-L1 or PD-L2, limiting immune activation. PD1-deficient mice exhibit hyperactivated immune responses and autoimmune diseases. Despite its predicted molecular weight, PD-1 often migrates higher in SDS-PAGE.
