Target Information

The ABL1 proto-oncogene encodes a cytoplasmic and nuclear protein tyrosine kinase involved in cell differentiation, division, adhesion, and stress response. The c-Abl protein’s activity is negatively regulated by its SH3 domain; deletion of this domain converts c-Abl into an oncogene. Its DNA-binding activity is regulated by CDC2-mediated phosphorylation, linking it to cell cycle control. In chronic myelogenous leukemia and some acute lymphoblastic leukemias, a (9;22) chromosomal translocation produces the Philadelphia chromosome, fusing c-Abl sequences from chromosome 9 with the Bcr gene on chromosome 22. The c-Abl oncogene was first identified as the viral transforming gene of Abelson murine leukemia virus (A-MuLV). c-ABL also plays a role in DNA repair by activating proapoptotic pathways when DNA damage is irreparable.
