Target Information

The ABL1 proto-oncogene encodes a cytoplasmic and nuclear protein tyrosine kinase involved in cell differentiation, division, adhesion, and stress response. The c-Abl protein’s activity is negatively regulated by its SH3 domain; deletion of this domain converts it into an oncogene. c-Abl’s DNA-binding activity is regulated by CDC2-mediated phosphorylation, indicating a role in the cell cycle.

In chronic myelogenous leukemia (CML) and some acute lymphoblastic leukemias (ALL), a (9;22) chromosomal translocation fuses c-Abl from chromosome 9 with the Bcr gene on chromosome 22, forming the Philadelphia chromosome. This fusion results in the oncogenic Bcr-Abl protein. The c-Abl oncogene was first identified as the viral transforming gene of Abelson murine leukemia virus (A-MuLV). Additionally, c-Abl may regulate DNA repair by activating pro-apoptotic pathways when DNA damage is irreparable.
