Target Information
Epithelial sodium channels (ENaC) are amiloride-sensitive members of the degenerin/epithelial sodium channel (Deg/ENaC) superfamily of ion channels.
These channels share structural similarity with two short intracellular termini, two membrane-spanning segments, and a large extracellular loop containing a conserved cysteine-rich region.
There are three homologous isoforms of ENaC: alpha, beta, and gamma.
ENaC in kidney, lung, and colon plays a crucial role in trans-epithelial sodium and fluid balance and mediates aldosterone-dependent sodium reabsorption in the distal nephron, regulating blood pressure.
ENaC is partially regulated through association with the CFTR chloride ion channel.
Gain-of-function mutations in beta or gamma ENaC can cause severe arterial hypertension (Liddle’s syndrome), while loss-of-function mutations in alpha or beta ENaC cause pseudohypoaldosteronism (PHA-1).
