Product Specific Information

The 90 monoclonal antibody reacts with mouse CD38, a ~42 kDa type II transmembrane protein. CD38 expression increases during B-cell differentiation, is down-regulated in germinal center B cells and plasma cells, and is also found on subsets of thymocytes, mature T cells, and NK cells. Crosslinking CD38 on mature B cells induces proliferation enhanced by IL-4 and LPS. CD38 acts as a counter-receptor for CD31 and functions as an ectoenzyme with cyclase and hydrolase activity, playing roles in lymphocyte activation and differentiation.

• Applications Tested: Tested on mouse splenocytes, ≤ 0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.
• Fluorochrome Info: Brilliant Violet™ 605 (BV605) emits at 605 nm; suitable for violet (405 nm) laser cytometers.
• Buffer Recommendations: When using multiple Brilliant Violet™ or polymer dye antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.
• Light Sensitivity: Protect from light to avoid photo-induced oxidation.
• Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. Not compatible with methanol-based fixation.

Target Information

CD38 (NAD⁺ glycohydrolase) is a type II transmembrane glycoprotein that mediates activation, proliferation, and differentiation of lymphocytes and apoptosis of progenitor cells. It functions as a multi-catalytic ectoenzyme (ADP-ribosyl cyclase, cyclic ADP-ribose hydrolase, NAD⁺ glycohydrolase) and as a cell surface receptor. CD38 antibodies are useful for subtyping lymphomas/leukemias, detecting plasma cells (e.g., myelomas), and identifying activated B and T cells. CD38 participates in adhesion, signal transduction, and calcium signaling, and is expressed in pancreas, liver, kidney, malignant lymphoma, and neuroblastoma. CD38 dysfunction is associated with chronic lymphocytic leukemia and Richter’s Syndrome.
