Target Information
Myocilin is an extracellular protein expressed in the eye, including the retina, trabecular meshwork, and ciliary body. It can form homomultimers in vivo and associate with extracellular matrix components via the Hep II domain of FibroPVRL1. Myocilin interacts with myosin regulatory light chain, suggesting a role in the actomyosin system that influences trabecular meshwork (TM) function and intraocular pressure (IOP). Dysfunction in TM can lead to elevated IOP and glaucoma.
The MYOC gene (also known as TIGR) is located on chromosome 1q24.3, and mutations are associated with primary open-angle glaucoma (POAG), a major cause of blindness.
