Target Information
Adrenergic receptors (ARs) are 7-transmembrane domain G-protein-coupled receptors that bind catecholamines such as epinephrine and norepinephrine.
Nine receptor subtypes have been identified: three alpha-1 ARs (1A/D, 1B, 1C), three alpha-2 ARs (2A, 2B, 2C), and three beta ARs (1, 2, 3).
ARs are involved in various physiological and pathological processes including:
- Hypertension
- Cardiac dysfunction (heart failure, ischemia, arrhythmia)
- Diabetes, glaucoma, depression, and impotence
Beta-1 adrenergic receptors (B1AR) play critical roles in cardiac function, development, and metabolism.
Knockout studies show that loss of the B1AR gene leads to prenatal lethality or abnormal cardiac function.
B1AR expression is regulated by thyroid hormones and the catecholamine-sensitive adenylyl cyclase system, with cAMP and CREM family transcription factors influencing its transcriptional control.
