Target Information
COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis and inflammatory prostaglandin production.
This conversion involves two reactions:
- Cyclooxygenase (COX) reaction: arachidonate → prostaglandin G2 (PGG2)
- Peroxidase reaction: PGG2 → prostaglandin H2 (PGH2)
COX2 is constitutively expressed in tissues such as endothelium, kidney, and brain, and up-regulated under pathological conditions including cancer and inflammation.
It is linked to increased cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis.
In cancer cells, COX2 promotes prostaglandin E2 (PGE2) production, affecting motility, proliferation, and apoptosis resistance.
COX2 activity can be inhibited by calcitriol (active Vitamin D) and glucocorticoids, and is targeted by NSAIDs like aspirin to reduce inflammation.
