Product Specific Information

The eBio4B10 monoclonal antibody reacts with mouse and human T-bet, a Th1-specific T-box transcription factor critical to the development of the Th1 CD4⁺ T cell lineage.
T-bet expression is induced by IFN-γ and regulates IFN-γ, IL-12Rβ, and IL-2 expression. It also plays a role in class-switch recombination in B cells.

Applications Reported:
For use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells.
Recommended ≤ 0.25 µg/test in 100 µL final volume (10⁵–10⁸ cells/test).
Optimize titration for best results.

Recommended Buffer Set:
Use the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and follow BestProtocols® Protocol B.

Target Information

T-bet (TBX21) is a T-cell specific transcription factor containing a T-box DNA-binding domain.
It regulates expression of the Th1 cytokine IFN-γ and contributes to Th1 lineage commitment.
T-bet cooperates with Stat4 in programming chromatin architecture for Th1 gene expression.
Genetic variations are associated with susceptibility to aspirin-induced asthma and nasal polyps.
