Product Specific Information
The KP1 antibody reacts with human CD68, a member of the sialomucin family related to lysosomal-associated membrane proteins (LAMPs) and scavenger receptors. CD68 is mainly an intracellular protein localized in late endosomes, with smaller amounts detected on the surface of myeloid-derived cells such as monocytes, macrophages, dendritic cells, neutrophils, basophils, mast cells, and myeloid progenitors. It may play roles in antigen processing/presentation and lysosomal membrane protection.
Expression has also been observed in activated T cells, a subset of B lymphocytes, and certain fibroblasts. The KP1 antibody has been reported for use in flow cytometry, immunoprecipitation, western blotting, and immunohistochemistry (both frozen and paraffin-embedded tissues).
Applications Tested:
Tested by IHC of formalin-fixed paraffin-embedded tissue using high pH antigen retrieval at 8 µg/mL. Optimal dilution should be determined experimentally.
Usage Note:
Centrifuge the conjugate solution briefly before use and use only the supernatant to minimize nonspecific background from aggregates.
Target Information
CD68 (Macrosialin) is a 110 kDa integral membrane glycoprotein expressed mainly on lysosomes of monocytes and macrophages, and to a lesser degree on dendritic cells and granulocytes. It plays a role in phagocytosis and lysosomal metabolism. CD68 expression is noted in interdigitating reticulum cells, histiocytic disorders, AML, and granulocytic sarcoma, and is elevated in CD34+ cells in several malignancies.
