Target Information
TSC2 (Tuberin, Tuberous sclerosis complex, TSC Complex Subunit 2)은 종양 억제 인자로 알려져 있으며, 소포 수송(vesicular transport), 세포 성장 정지(cell growth arrest) 조절, 스테로이드 수용체 매개 전사 조절에 관여합니다.
TSC2는 hamartin과 세포질 복합체를 형성하여 hamartin의 샤페론 역할을 할 수 있습니다. Ser939 및 Thr1462 부위의 인산화는 hamartin과의 상호작용을 조절하며, 이는 phosphoinositide 3-kinase/Akt 경로를 통해 다양한 성장 인자에 의해 자극됩니다.
TSC2는 RAP1A 및 RAB5 단백질의 내재적 GTPase 활성을 자극하여 세포 성장 조절에 기여할 수 있습니다. TSC2의 돌연변이는 RAP1A의 지속적 활성화를 유도하며, lymphangioleiomyomatosis 및 TSC2 angiomyolipoma와 같은 질환과 관련이 있습니다.
대체 스플라이싱을 통해 세 가지 서로 다른 isoform의 전사 변이가 생성됩니다.
