Target Information
DLAT encodes component E2 of the multi-enzyme pyruvate dehydrogenase complex (PDC), located in the inner mitochondrial membrane. PDC catalyzes the conversion of pyruvate to acetyl coenzyme A. The DLAT gene product, dihydrolipoamide acetyltransferase, transfers acetyl groups to coenzyme A. This protein is the antigen for antimitochondrial antibodies, which are present in nearly 95% of patients with primary biliary cirrhosis (PBC). In PBC, abnormal distribution and overexpression of DLAT in bile duct epithelial cells lead to immune-mediated destruction, cirrhosis, and liver failure.
