Target Information

The G-protein-coupled receptor (GPCR) superfamily is comprised of an estimated 600–1,000 members and represents the largest known class of molecular targets with proven therapeutic value.
GPR26 is a brain-specific orphan GPCR with constitutive activity mediated by G(s)-alpha protein, stimulating adenylate cyclase and elevating intracellular cAMP levels.
In the CNS, GPR26 is highly expressed in the amygdala, hippocampus, and thalamus, with weak expression in testis. It is down-regulated in glioblastoma.
Research has shown that depletion of GPR26 increases fat storage in C. elegans, and deficiency in the hypothalamus correlates with genetic susceptibility to obesity in mice.
GPR26 deficiency also leads to metabolic complications such as glucose intolerance, hyperinsulinemia, and dyslipidemia.
