Target Information
FOXO1 (FKHR, Forkhead Box O1)는 전사 인자(forkhead homeotic gene family)의 하위 패밀리에 속합니다. FOXO1은 핵 수용체 활성의 공동활성자(coactivator) 또는 공동억제자(corepressor)로 작용할 수 있으며, FKHR 단백질의 카복실 말단 영역에 존재하는 LXXLL 모티프를 통해 매개됩니다.
FOXO1과 PAX3의 결합은 alveolar rhabdomyosarcoma와 관련이 있으며, 항종양 활성, 자가포식(autophagy), 근육 성장 및 분화 과정과도 연관되어 있습니다.
FOXO1은 인슐린 신호전달의 주요 타깃으로, 산화 스트레스에 대한 대사 항상성을 조절합니다. 또한 FOXO1은 인슐린 반응 요소(IRE, 5-TT[G/A]TTTTG-3) 및 Daf-16 결합 요소(DBE, 5-TT[G/A]TTTAC-3)에 결합합니다.
이 단백질은 산화환원 균형, 조골세포 수, 골량 조절, 골격의 내분비 기능, 포도당 대사 조절, 그리고 RUNX2 전사활성 억제에 관여합니다. 간세포에서는 PPARGC1A와 함께 작용하여 IGFBP1, G6PC, PCK1 등의 유전자 발현을 촉진함으로써 포도당신생합성을 유도합니다. 또한, FOXO1은 CDK1, PKB/AKT1, SKT4/MST1 하위에서 세포 사멸을 조절하는 중요한 인자입니다.
