Target Information
FOXO1 (FKHR, Forkhead Box O1) is a transcription factor belonging to the forkhead homeotic gene family. It can function as a coactivator or corepressor of nuclear receptor activity via the LXXLL motif in its C-terminal region.
FOXO1 is implicated in alveolar rhabdomyosarcoma through its association with PAX3. It plays roles in anti-tumor activity, autophagy, and myogenic differentiation.
As the main target of insulin signaling, FOXO1 regulates metabolic homeostasis under oxidative stress and binds to the insulin response element (IRE) and Daf-16 family binding element (DBE).
FOXO1 regulates redox balance, osteoblast numbers, and bone mass, and modulates endocrine functions related to glucose metabolism. In hepatocytes, it promotes gluconeogenesis by activating genes such as IGFBP1, G6PC, and PPCK1.
