Target Information
Immunophilins are a family of soluble cytosolic receptors that bind to immunosuppressant agents such as cyclosporin A (CsA) or FK506.
- Proteins binding FK506 are termed FKBP (FK506 Binding Proteins).
- Proteins binding CsA are called cyclophilins (CyP).
Both CyP:CsA and FKBP:FK506 complexes inhibit calcineurin, a calcium- and calmodulin-dependent protein phosphatase involved in T-cell activation, explaining the immunosuppressive effects of CsA and FK506.
Immunophilins act as peptidyl prolyl cis-trans-isomerases (PPIase), catalyzing protein folding and rearrangement steps, which are inhibited by their respective immunosuppressants.
