Target Information
SMAD2 (MADH2, MAD2) regulates multiple cellular processes such as cell proliferation, apoptosis, and differentiation. It interacts with TGF-beta receptors via the SMAD anchor for receptor activation, playing a central role in TGF-beta signaling.
Phosphorylation by ERK1 enhances transcriptional activity, while calmodulin negatively regulates it. Upon TGF-beta stimulation, SMAD2 is phosphorylated, dissociates from SARA, and associates with SMAD4 to translocate into the nucleus, forming transcriptional repressor complexes.
SMAD2 can also be phosphorylated by activin type 1 receptor kinase, mediating activin signaling. Alternatively spliced transcript variants encoding SMAD2 have been observed.
