Target Information
CD180 (also known as RP105, Bgp95, LY64) is a type I membrane glycoprotein of the Toll-like receptor (TLR) family. Unlike other TLRs, CD180 lacks the TIR domain in its cytoplasmic tail. Its expression depends on coexpression with MD-1 and parallels that of TLR4 on antigen-presenting cells. CD180 modulates LPS recognition and B cell signaling by interacting with the TLR4 complex, inhibiting ligand binding. Engagement of CD180 by monoclonal antibodies triggers B cell activation, upregulation of CD80/CD86, and cell enlargement. The extracellular domain contains leucine-rich repeats (LRR) forming the RP105/MD-1 receptor complex. CD180 dysfunction is linked to Kawasaki Disease and may contribute to rheumatic disease pathology.
