Product Specific Information

This antibody detects endogenous levels of Progesterone Receptor only when phosphorylated at Serine 400.

Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.
PR exists as two isoforms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), generated by alternative translation initiation sites.
PR-B is the transcriptionally active form responsible for activating genes related to endometrial maintenance, pregnancy, and inhibition of ovulation.
PR-A lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon binding progesterone, conformational change allows receptor dimerization and binding to progesterone response elements (PRE), initiating transcription.
A null mutation in the PGR gene results in reproductive abnormalities.
