Product Specific Information

Description:
The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha has cytolytic activity against a range of tumor cells and is important in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form. The MAb11 antibody is a neutralizing antibody.

Applications Reported:
This antibody has been reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells.
Use at 5 µL (0.5 µg) per test (final volume 100 µL).
Cell number can range from 10⁵ to 10⁸ cells/test.

Excitation/Emission:
488 nm / 519 nm; Laser: Blue Laser.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers and shows a single 17 kDa band on SDS-PAGE under non-reducing conditions. Knockout studies in mice suggest neuroprotective functions and tumor necrosis induction. TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
