Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with pregnancy establishment and maintenance.
PR exists as two forms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), produced via alternative translation initiation sites.
PR-B is transcriptionally active, regulating genes involved in endometrial maintenance, pregnancy, and ovulation inhibition.
PR-A differs from PR-B by a 165 amino acid deletion at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.
Inactive PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes conformational change, dimerizes, and binds to progesterone response elements (PRE), activating transcription.
A null mutation in the PGR gene can lead to reproductive abnormalities.
