Product Specific Information

  • Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10–20 minutes, followed by cooling at room temperature for 20 minutes.
  • After retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
  • Suggested positive controls: skin, squamous cell carcinoma, or prostate tissue.

Target Information

Cytokeratins (CK) are intermediate filaments of epithelial cells, found in both keratinizing tissues (e.g., skin) and non-keratinizing cells (e.g., mesothelial cells). At least 20 different CKs have been identified and are classified into two main types:

  • Type I (acidic polypeptides)
  • Type II (basic polypeptides)

Keratin 14 belongs to the type I subfamily of low molecular weight keratins and typically pairs with keratin 5. It helps distinguish stratified epithelial cells from simple epithelial cells and is useful in identifying squamous cell carcinomas. Keratin 14 is also considered a prognostic marker in breast carcinomas. Both acidic and basic keratin family members are expressed in all epithelial cells.
