Target Information

Sonic Hedgehog (SHH) is expressed only during embryogenesis and plays a key role in patterning the early embryo. It is crucial for ventral neural tube formation, anterior-posterior limb axis development, and ventral somite patterning. The SHH protein is produced as a precursor that undergoes autocatalytic cleavage. The N-terminal fragment is soluble and responsible for signaling activity, while the C-terminal fragment aids in precursor processing and attaches a cholesterol moiety to the N-terminal product. This modification restricts the N-terminal product to the cell surface, preventing diffusion throughout the embryo. Defects in SHH or its signaling pathway can lead to holoprosencephaly (HPE) and may be associated with VACTERL syndrome.
